Abstract

Objective By measuring and comparing the relaxant and contractile abilities of lung bronchial smooth muscle and the epithelial dependent relaxation (EpiDR) ability between heart-beating donor (HBD) and non-beart-beating-1 h donor (NHBD-1 h) ,to explore the feasibility of the NHBD-1 h lung to be used for lung transplantation. Methods Sixteen Swedish pigs were divided into two groups ran-domly: HBD group and NHBD-1 h group. Several bronchial rings were harvested from these pigs with inner diameters of all about 1.5 mm and about 1.5 mm long without branches, and then placed into the organ bath device. The reactions of the rings to some certain drugs [acetylcholine (Ach) ,arachidonic acid natri-um,etc.] were tested, and the relaxant and contractile abilities of lung bronchial smooth muscle and EpiDR ability analyzed. Resulls Between HBD group and NHBD-1 h group, there was no significant difference in contraction of bronchial smooth muscle caused by acetylcholine (5.18±0.07 vs 5.10± 0.11) (P > 0.05 ) ; bronchial smooth muscles in both groups could were completely relaxed by papaverin hydrochloride; after removing the spontaneous relaxation interference in HBD and NHBD-1 h groups (HBD-B:5.18±0.10 vs 5.20±0. 11 ,P >0.05 ;NHBD-1 h-B:5.10±0.06 vs 5.11±0.07,P >0.05), there was no significant differences in average EpiDR ability ( 1.26 ± 0.05 vs 1.23 ± 0.07 ) between two groups ( P > 0.05 ) also. Conclusion After warm ischemia for 1 h, the relaxant and contractile abilities of lung bronchial smooth muscle keep intact, and the epithelial dependent adjustment of lung bronchi shows no significant difference between HBD and NHBD-1 h.
