Abstract
Human growth hormone (hGH) isohormones B and E were isolated by preparative polyaerylamide gel electrophoresis; two other fractions containing predominantly the 2 isohormones (B and C), (D and E), were prepared. Specific activities of the 4 preparations containing isohormones B, BC, DE and E were determined in the rat tibial-line bioassay for hGH; the pigeon crop-sac bioassay for prolactin; and in a radioimmunoassay for hGH. The immunological activity of the 4 preparations was the same. Tibial-line bioactivity increased progressively from hGH-B to hGH-E by a total factor of 8. Prolactin bioactivity increased progressively from isohormone B to E by a total factor of 3. Since isohormone E can be generated from isohormone B by digestion with plasmin, these findings may potentially increase the pool of available hGH biological activity. (Endocrinology 93: 866, 1973)
