Abstract

Glycyrrhiza uralensis (licorice) is one of the most frequently prescribed agents in traditional herbal medicinal. We have previously observed that hexane/ethanol extract of Glycyrrhiza uralensis (HEGU) induced apoptosis and cell cycle arrest in DU145 human prostate cancer cell growth. This study examined the effects of the active compound isoangustone A in HEGU on apoptosis of DU145 cells. Isoangustone A (0 ‐ 7.5 mg/L) dose‐dependently reduced the viable cell numbers and increased apoptotic cell numbers in DU145 cells. Western blot analysis of total cell lysates revealed that isoangustone A increased the levels of Fas, TRAIL, cleaved caspase‐8 and Mcl‐1S and decreased the levels of Mcl‐1L. In addition, isoangustone A dose‐dependently increased the cleavage of poly(ADP‐ribose) polymerase and caspases 9, 7, and 3. We have demonstrated that isoangustone A inhibits DU145 cell growth via the induction of apoptosis.
