Abstract

DNA is extracted from purified mitochondria of potato tubers. This DNA cannot be resolved into two strands by alkaline CsCl gradients in our experiments. Poly (G), poly (U) and poly (I,G) interact with the plant mitochondrial DNA as shown by the marked shift in buoyant density that they produce on denatured DNA. Poly (A) and poly (C) do not lead to detectable interactions in standard conditions whereas a slight fixation of poly (C) occurs at acidic pH. These results suggest that the plant mitochondrial DNA contains d-A and d-C rich clusters and, in a lesser extent, d-G rich clusters.
