Abstract

Objective To observe the effects of antisense RhoC gene on the expression of matrix metalloproteinase-2 (MMP-2) in QBC939 cells.Methods Antisense RhoC cDNA fragment eukaryotic expression vector was transfected into QBC939 cells.Reverse transcription-polymerase chain reaction ( RTPCR) and Western blotting were used to examine the expression of MMP-2 in vitro.Results There was no significant difference in absorbance (A) values of the expression of MMP-2 mRNA between QBC939 cells (0.588 ± 0.074) and QBC939-V cells ( 0.629 ± 0.061 ),P > 0.05,but those in QBC939-As cells (0.286 ± 0.097) were lower than in QBC939 cells and QBC939-V cells ( P < 0.05 ).There was no significant difference in the expression of MMP-2 expression between QBC939-AS group and control group (P <O.05).The expression levels of MMP-2 protein and mRNA in QBC939-AS group were significantly lower than in control group.Conclusion Antisense RhoC could inhibit the expression of MMP-2 gene in 0BC939 cells.
