Abstract

Obesity is characterized at the cellular level by an increase in the number and size of adipocytes differentiated from fibroblastic preadipocytes in adipose tissue. Inhibition of adipocyte differentiation is one approach among the anti‐obesity strategies. Corni fructus (SSY) has been used as a folk medicine in Korea. In the present study, we investigated the molecular mechanisms of the anti‐obesity effect of SSY water extract on adipocyte differentiation of 3T3‐L1 cells. The differentiation of 3T3‐L1 preadipocytes cells was induced by hormone cocktail, including dexamethasone, isobutylmethylxanthine (IBMX), and insulin. Effects of SSY treatment on adipocyte differentiation were measured by Oil Red O staining, triglyceride and cell viability assays. In addition, the protein expression of peroxisome proliferation activated receptor‐γ (PPAR‐γ) and AMPK‐activated protein kinase (AMPK) was detected by western blotting. 1000 ppm SSY water extract significantly reduced cell number but decrease in cell number was not due to a cytotoxic effect, because cytotoxicity appeared at greater than 2000 ppm. Triglyceride accumulation and PPAR‐γ expression were decreased by SSY water extract in a dose‐dependent manner but failed to show increase of AMPK in SSY treated cells. These data suggest that the anti‐obesity effect of SSY is mediated by the PPAR‐γ signaling pathways in 3T3‐L1 preadipocytes during differentiation.
