Abstract

Uranyl or ferric ions in the growth medium of Acanthamoeba rendered these nuclei Feulgen negative and ferric ions in the growth medium of Tetrahymena greatly reduced Feulgen staining. A similar inhibitory effect was produced when fixed Acanthamoeba or Tetrahymena cells were exposed to the same ions either before or after acid hydrolysis but not after staining with Schiffs, reagent. A mechanism to account for this inhibition has been proposed. The importance of these findings to workers performing quantitative Feulgen reactions has been discussed.
