Abstract

Penicillamine (30 mg⁄kg⁄day orally for 4 weeks) increased neutral-salt-soluble collagen, while 17β-estradiol cypionate (50–100 Mg⁄kg⁄wk×6) decreased both neutral-salt- and acid-soluble collagen in guinea pig skin. When penicillamine was administered during the last 4 weeks of a 6-week course of 17β-estradiol cypionate, the increase in collagen solubility produced with penicillamine alone did not occur. Penicillamine had no significant effect on collagen solubility in the carrageenan granuloma, while 17β-estradiol cypionate lowered neutral-salt and acid-soluble collagen. The effects of 17β-estradiol cypionate on guinea pig skin and granuloma soluble collagen, skin, bone and aorta total collagen, and on the in vivo incorporation of administered 14C-L-proline into collagen of these tissues were not modified by the coincident administration of penicillamine. A low-calcium diet in rats reduced total bone mass and in vitro collagen biosynthesis by metaphysial bone fragments; penicillamine administered during th...
