Abstract
The effects of actinomycin-D and puromycin on RNA and protein synthesis were studied in newborn rat epidermis. These antibiotics inhibited RNA and protein synthesis and resulted in ultrastructural changes of keratohyalin granules and of other granular cell constituents: Actinomycin-D produced Medusa bead-like keratohyalin granules and puromycin prevented enlargement of keratohyalin granules by interfering with the insertion of tonofibrils and the addition of dense deposits. This work suggests that the dense deposits observed on keratohyalin granules may originate in the nucleus.
