Abstract
Objective To observe the inhibiting effect of down-regulation of integrin-pl expression on hepatic metastasis of human colon carcinoma cell (HT-29). Methods Integrin-pl antisense oligodeoxynucleotide (ASODN) was transfected into HT-29 cell by liposome. The expressions of integrin-β1 mRNA andProtein were determined by RT-PCR and Western-blot. The migration of HT-29 cells was assayed using transwell cell culture chambers in vitro. The models of hepatic metastasis in nude mice were established by the intrasplenic injection of transfected HT-29 cells. Thirty days later,the number and size of hepatic metastases were calculated,and the expression of integrin-β1 was studied by immunohistochemistry (SP). Results In contrast with the control group,the expression of integrin-β1 mRNA andProtein were significantly down-regulated in the experimental group (P<0. 05);In vitro,the number of migrated HT-29 cells in the experimental group was far less than that of the control group (P<0. 05);In vivo,the average number (4.00±0.93)/mouse,volume (10.10±6.50) mm~3/mouse,weight (0.0440±0.0008) g/mouse andPositive cellPercentage (28.6±3.79) % of hepatic metastases of the experimental group in nude mice were markedly decreased compared with the control group (P<0. 05 ). Conclusion Downregulation of integrin-βl expression mayPlay a key role in inhibiting hepatic metastasis of human colon carcinoma. Key words: Colon carcinoma; Hepatic metastasis; Integrin-β1
