Abstract
Objective To observe the influence of RNAi against lysyl oxidase (LOX) gene on biological features of MDA-MB-231 cells.Methods The MDA-MB-231 cells were transfected with lentiviral interference vector (LOX-RNAi-LV),and divided into three groups:the experimental group,the blank group and the negative group.The growth and proliferation of cells were determined by using methyl thiazol tetrazolium (MTT) assay,cell colony formation and flow cytometry.The migration and invasion were observed by using transwell assays.Results The cell growth rate in the experimental group,the blank group and the negative group at 24,48,72 and 96 h was (12.73 ± 3.30) %,(33.93 ± 1.99) %,(52.63 ±1.04)%,(63.80±0.70)%;0,0,0,0; (7.60±6.27)%,(10.30±1.49)%,(9.10±2.69)%,(6.03 ± 1.21) %,respectively (P < 0.05).Cells invading between three groups was 47 ± 2 ; 100 ± 1 ; 88 ± 2 (P < 0.05).Cell migration was 63 ± 2 ; 118 ± 2 ; 96 ± 2 (P < 0.05).The difference of the cell cycle was not statistically significant (P > 0.05).Conclusion After the LOX gene expression restrained,the creature feature of MDA-MB-231 were effectively suppressed.
