Abstract
Chemostat culture for non-adhesive recombinant CHO cells (rCHO) at the wide range of glucose and glutamine feed concentration was carried out in the serum-free media. The specific monoclonal antibody production rate was independent of glucose concentration, however increased by decrease of glutamine concentration in low glutamine concentration range. Intracellular fluxes were estimated from extracellular utilization and production rates. It was elucidated that in low glutamine concentration, fluxes of glycolysis and the TCA cycle were accelerated, and the flux of the direct pathway to α-ketoglutalate increased with decrease of glucose concentration, however the flux of glutamine decomposition was not influenced by glucose concentration.
