Abstract

Objective To investigate the expression of Tie 2 gene induced by inflammation cytokines TNF-α and IL-1β in vascular endothelium cells and explore the mechanism of angiogenesis under inflammation. Methods mRNA and protein expressions of Tie 2 were investigated by quantitative PCR and Western blot in primary human aortic endothelial cells (HAECS) and/or primary human pulmonary artery endothelial cells (HPAECS)induced by inflammatory cytokines. Results TNF-α and IL-1β could increased the expression of Tie 2 in primary endothelial cells. The expression of Tie 2 was significantly increased 24 hours after stimulation of TNF-α in HAECS and HPAECS and IL-1β in HPAECS. in HPAECS, the effect of both arrived the peak at 36 hours after stimulation. Conclusion The expression of Tie 2 could be increased by inflammatory cytokines TNF-α and IL-1β in human vascular endothelial cells. It possibly contributes for angiogenesis under inflammation.
