Abstract

Somatic embryogenesis is useful for preservation, clonal multiplication and genetic manipulation. The present study established a somatic embryogenesis system of three mango genotypes: 'Baixiangya', a polyembryonic cultivar; 'Tainong 1', a monoembryonic cultivar and 'Guifei', Immature fruits were collected from these cultivars 40-60 days after anthesis in the Germplasm Research Institute of Tropical Crops of the Chinese Academy of Tropical Agriculture Sciences, Hainan, China during February and March. The ovules were aseptically removed from the immature seeds and cultured on the modified B5 medium with 2.74 mM L-glutamine, 0.57 mM ascorbic acid, 6% (w/v) sucrose and 0.8% (w/v) agar powder. Induction of embryogenic competence was controlled by genotype and various growth regulators (2,4D or 6-BA alone, or 2,4D with 6-BA). Explants grown on the basal medium (control) were also inductive but at a very low efficiency (<10%). All treatments were significantly inductive compared with the control. 'Baixiangya' cultivar had the highest embryogenic potential, followed by 'Guifei' and 'Tainong 1'. The most effective culture was 'Baixiangya' cultivar treated with 2,4-D 5 mg/l + 6-BA 2 mg/l.
