Abstract

A novel cellulase inducer named MASS was isolated from molasses alcohol stillage (MAS). HPLC analysis identified it as a novel hetero-oligosaccharide composed of the monosaccharides glucose, fructose and xylose in the ratio of 6:3:1; size-exclusion chromatography determined its molecular weight about 4,200 Da. MASS induced both the gene transcription and enzyme activities of cellobiohydrolase (EC 3.2.1.91) (CBH) in the cellulolytic fungus Trichoderma reesei. Cellulase activity assays showed that MASS induced a much stronger CBH activity than cellobiose and lactose, but a weaker one than sophorose. Real-time PCR revealed that MASS and other inducers regulate cbh gene transcription and the induction of gene transcription was correlated with the change of protein activities.
