Abstract

Soeka YS. 2015. The characterization and identification chitinase enzyme in actinomycetes KRC 21.D isolate derived from Cibodas Botanical Garden. Pros Sem Nas Masy Biodiv Indon 1: 1156-1161. The aim of the research was to know the activity of chitinase enzyme in actinomycetes collected from Cibodas Botanical Garden. Based on the appearance of the clear zone around colonies on medium containing 1% colloidal chitin substrate, isolates were selected qualitatively. The enzyme activity was measured by spectrophotometer at I» 540 nm and examined at different incubation period, pH, temperature as well as 1mM solution of different divalent and monovalent cations. The results of the research showed that the highest chitinase activity, 7.2 A— 10-3 U/mL, was found at 7 days incubation, while the chitinase activity on growth medium at pH 10 and at 25°C was 8.05A—10-2 U/mL. Experimental result of influencing metal ions showed that Na+1 ,Ca2+, Zn2+, Mn2+, Co2+ ions act as an enhancer while Hg2+ ions acts as inhibitors of chitinase enzyme activity. The molecular identification revealed that the isolate actinomycetes KRC 21.D was Streptomyces macrosporeus.
