Abstract

Abnormal calcium (Ca) leak via cardiac ryanodine receptor 2 (RyR2) is enhanced by high extracellular [glucose] via O-GlcNAcylation of CaMKII and may be important in diabetic cardiomyopathy (PMID: 24077098). Indeed, CaMKII O-GlcNAcylation and activity are elevated in humans and animal diabetic models. Here, we tested whether CaMKIIδ and the site identified as an O-GlcNAcylation target (Ser280) are required for promoting SR Ca leak. We measured Ca sparks, Ca transients, and sarcoplasmic reticulum (SR) Ca content in isolated mouse ventricular cardiomyocytes.
