Abstract
Background:Calpain has been associated with the pathophysiology of Alzheimer‘s disease (AD) and with apoptotic neuronal cell death.Methods: A total of 64 patients with AD and 52 ageand gender-matched cognitively healthy elderly controls were included in the study. We measured c alpain activity and levels of Ab1-42, h-tau and p-tau.Results:AD patients showed significantly increased calpain activity in CSF and significantly decreased calpain activity in serum and plasma compared with healthy controls. In addition, calpain activity in CSF was significantly inversely correlated with Ab1-42 CSF levels and positively correlated with p-tau CSF levels in AD patients and healthy controls. Conclusions: This is the first study showing increased calpain activity in CSF of AD patients. Calpain activity could represent a novel diagnostic biomarker and promising treatment target for AD.
