Abstract

Adrenomedullin (ADM) is a tissue protecting peptide hormone and hormone production and plasma levels increase when kidneys are subjected to hypoxia in vivo. We examined the expression of ADM in the different regions of the rat kidney in response to uni- and bilateral ureteral obstruction (UUO and BUO), conditions known to be characterized by hypoxia in the renal tissue. Rats were subjected to either 24h UUO, BUO or sham-operation (SHAM). Kidneys were then removed and dissected into zones: Cortex+outer stripe of outer medulla (C+OSOM), inner stripe of outer medulla (ISOM) and inner medulla (IM) and prepared for RNA extraction. In a subset of animals kidneys were perfusion fixed for immunohistochemistry (IHC). BUO significantly increased ADM mRNA levels 3 fold in C+OSOM (P<0.01), 4 fold in ISOM (p<0.01) and 7 fold in IM (p<0.05) compared to levels in SHAM. IHC showed enhanced ADM labelling in proximal tubules, thick ascending limbs, distal convoluted tubules, and collecting ducts compared to SHAM. Immunolabeling was not detected in loops of Henle or glomeruli. In the obstructed kidney from UUO rats ADM levels only increased in IM (> 5 fold, p<0.05) compared to SHAM. In conclusion ADM mRNA and ADM immunolabeling increases in kidneys subjected to ureteral obstruction suggesting that ADM may play a protective role in UUO and BUO.
