Abstract

Kayu kuku (Pericopsis mooniana Becc. ex. Heyne) is one of wood elite species that have high economic value. Seed germination and shoot multiplication of kayu kuku, an endangered tree species in Indonesia, have been studied using in vitro technique. Seeds collected from Riau province were used as material whereas the Murashige and Skoog (MS) medium as basal medium. Sterile seeds were germinated in MS medium without plant growth regulator. The cotyledone nodes of the 7 weeks seedling produced from seed germination were used as explant for shoot multiplication. Five treatments concentration of each Benzyl Adenine (BA) (0.75 mg/l – 3.0 mg/l) and thidiazuron (0.1 – 1.5 mg/l) with three replications were used as media for shoot multiplication. The study showed that 100% of seed germinated in MS medium with no plant growth regulator. The treatment of MS+BA 0.75 mg/l showed the best medium treatment for shoot multiplication as indicated with the highest number of shoot and its total height (2.3 shoots and 6.8 cm in average). The MS medium+BA 0.75 mg/l was also the best visual shoot performances at 12 weeks old observation.
