Abstract

The present study has examined the effect of Day-7 and Day-8 (Day 0 = estrus) bovine uterine flushings on the in vitro development of mouse embryos at the two-cell stage and Day-7 and Day-8 bovine embryos. The uterine fluids (protein concentrations ranged from 35 to 250 μg/ml) were collected by flushing the uterus nonsurgically with Ham's-F10 medium. The results showed that 93.3% bovine embryos cultured in bovine uterine secretions in the absence of paraffin oil for 24 h presented signs of degeneration or did not develop, whereas 94.7% embryos (bovine and mouse) cultured in bovine uterine secretions under paraffin oil did develop. This study reveals the presence of a substance in Day-7 and Day-8 bovine uterine flushes that inhibited the in vitro development of bovine embryos in absence of paraffin oil but allowed the development of mouse and bovine embryos in the presence of paraffin oil. Thus, when bovine embryos are collected for transfer, they need to be removed as soon as possible from the uterine flushings after collection in order to remove them from a hostile environment.
