Abstract

Callus induction and regeneration of whole plants of the tree legume Dalbergia latifolia Roxb. (Indian Rosewood) was achieved in tissue culture under defined conditions. Callus was initiated from shoot and leaf segments and maintained on Murashige and Skoog (MS) basal medium that had both an auxin(s) and a cytokinin (kinetin). A compact and green callus that ultimately developed shoot buds was obtained on MS medium containing naphthaleneacetic acid (NAA) (3 mg/1) and benzylaminopurine (BA) (1 mg/1) in combination. Callus survived several subcultures. With an increased concentration of BA (3 to 5 mg/1) and a decreased concentration of NAA (0.5 mg/1) in the medium, 10 to 12 shoot buds appeared on the callus. Bud initiation, however, was effective with BA at 5 mg/1 and NAA at 0.5 mg/1. Buds developed well and elongated into shoots in BA at 3 mg/1 and NAA at 0.5 mg/1. Shoots were separated and rooted on half-strength MS medium with indolebutyric acid (1 mg/1).
