Abstract

Summary Polyethylenimine-poly(hydroxyethyl glutamine) copolymer (PEI-PHEG) could effectively form positively charged nanoparticles with pDNA. The PEI-PHEG copolymer exhibited much lower cytotoxicity and higher gene transfection efficiency than PEI-25 k with different cell lines in vitro. PEI-PHEG/pDNA complex demonstrated excellent transfection efficiency in animal tests. PEI-PHEG could efficiently deliver siRNA to cells to silence the target gene. It could also complex vascular endothelial growth factor siRNA (VEGF siRNA) to achieve the aim of tumor therapy.
