Abstract

Cyclin-dependent kinase 5(cdk5) is a tightly regulated protein that is involved in a variety of neuronal functions. The hyperactivity of cdk5 associated with p25 a cleaved complex of its activator protein p35 is believed to be the causative factor of Alzheimer’s disease (AD) and other neurodegenerative diseases. Many drugs that are available in the market such as Roscovitine Aminothiazole target the ATP binding pocket of cdk5 and inhibit its activity. As a result, there are potential side effects due to non-selective inhibition of both the endogenous cdk5-p35 and the pathological cdk5-p25 complex.
