Abstract

Objective To elucidate the expression of costimulatory molecule B7-1 and B7-2 in liver hypothermic irrigation and reperfusion model of rats and its significance.Methods Thirty rats were randomly divided into three groups:group A,hypothermic irrigation for 20 min; group B,hypothermic irrigation for 30 min; and group C,sham operation.The expression of B7-1 and B7-2 mRNA in the livers was detected by using real-time reverse transcription polymerase chain reaction (RT-PCR).Results The B7-1 and B7-2 expression levels were at very low levels in the liver tissues of group C (0.226 ± 0.053,and 0.339 ±0.049),but the mRNA expression levels of B7-1 and B7-2 were significantly enhanced in group A or group B (P<0.01).In addition,both B7-1 and B7-2 levels were higher in group B than in group A (B7-1:0.475± 0.079 vs 0.367 ± 0.052 ; B7-2:0.666 ± 0.039 vs 0.595 ± 0.050 ; P < 0.01).Conclusion Hypothermic irrigation and reperfusion may increase the immunogenticity of the liver by upregulating the costimulatory molecules B7-1 and B7-2.
