Abstract

In this paper the process of biosynthesis of fungal laccases is addressed with the investigation of methods for improvement of the enzyme’s production efficiency and prolongation of the culture’s performance. The research is focused on improving the biosynthesis results through repeated discontinuous fed-batch cultures, and in situ integration of the process with the first step of downstream processing – biomass separation via specially constructed rotating filter. Four different strategies were applied with varied frequency of feeding and whether the filter was used or not. Based on obtained results, it is claimed that biosynthesis of laccase conducted in repeated discontinuous fed-batch mode provides satisfactory improvement in terms of laccase production per run. The total laccase recovery in the process with daily refills was more than 6 times higher than in the batch mode performed in the same bioreactor.
