Abstract
IT is now firmly established that antibody production, delayed hypersensitivity to many antigens and the production of experimental auto-immune diseases are all enhanced by the use of Freund's adjuvant. During the past year I have made several unsuccessful attempts to induce auto-immune lesions in guinea pigs by inoculation of all four foot-pads with homologous and heterologous tissue antigens (lymph nodes, cartilage, stomach mucosa, blood vessel, etc.) incorporated in complete Freund's adjuvant. However, a striking featuie in all animals was the enlargement and induration of regional lymph nodes, with disruption of their architecture, and proliferation of macrophages and epithelioid cells. Impressed by those changes in lymph nodes, and taking into account that lymph nodes play an important part in immune mechanisms, it was thought that such animals might be “immunologically impaired”. They consequently might not be able to develop adequate delayed hypersensitive responses to an antigen later injected.
