SFORCIN1 1 Universidade Estadual Júlio de Mesquita Filho - UNESP, Departamento de Microbiologia e Imunologia, Brazil Introduction: Propolis is a natural product elaborated from the material collected by honeybees from different plants. This product shows a complex chemical composition which varies according to the botanical sources. Some studies associate the biological activity of propolis to its isolated compounds, however, little is known concerning the combination between its compounds that could lead to a similar action exerted by propopolis. This work aimed to investigate whether the combinations between three phenolic acids (caffeic, cinnamic and p-coumaric acids) are responsible for immunomodulatory effects of propolis on human monocytes. Methods and Results: Human monocytes were obtained from 10 healthy donors and treated with isolated compounds and their combinations (cafeic+cinnamic; caffeic+p-coumaric; cinnamic+p-coumaric; and caffeic+cinnamic+p-coumaric), according to their concentrations in propolis composition (20 µg/mL), simultaneously with LPS, for 18h. After this period, the cell viability was assessed by colorimetric MTT assay. After treatments, supernatants were harvested and the production of TNF-α and IL-10 was evaluated by enzyme-linked immunosorbent assay (ELISA). Hydrogen peroxide (H2O2) release by human monocytes was determined according as well. Significant differences between treatments were determined by analysis of variance (ANOVA), followed by Dunnett’s test (P<0.05). In comparison to propolis, isolated acids and their combinations maintained the LPS-induced TNF-α production, without affecting monocytes viability. On the other hand, cinnamic acid increased IL-10 production (693.6 ± 94.9 pg/mL). Regarding to H2O2 production, cinnamic (3.7 ± 0.6 nM) and p-coumaric (5.2 ± 1.0 nM) acids, and the combination caffeic+cinnamic (5.1 ± 0.9 nM) showed a similar production to that displayed by propolis (5.5 ± 0.3 nM). Conclusion: Data suggest that the compounds are related to the biological activities of propolis, and these effects may change according to different combinations between the phenolic acids. Nevertheless, further studies about the interaction of propolis compounds are still necessary.
