Abstract

Abstract A non-radioactive digoxigenin labelled DNA probe specific for Erwinia carotovora subsp. atroseptica, the causal agent of potato blackleg, has been developed. The probe is a randomly cloned 2100 bp fragment from a genomic library of one E. carotovora subsp. atroseptica strain. In a dot-blot DNA hybridization system, the probe was tested against a collection of 70 E. carotovora strains from various geographical regions, and of numerous serogroups. The probe reacted positively with 38 out of 40 E. carotovora subsp. atroseptica strains, but not with any of the 30 E. carotovora subsp. carotovora strains. Additionally, the probe did not react with 15 other related bacteria, saprophytic bacteria from soil and water, or other potato pathogenic bacteria. The detection limit of the probe is 1–10 ng total genomic DNA.
