Abstract

To identify new cry 1-type crystal protein genes, 52 Bacillus thuringiensis isolates which produced bipyramidal shaped crystal inclusions were selected. PCR-RFLP analysis of 52 Bt isolates revealed 6 distinct patterns, and 6 standard isolates were selected from each RFLP group. Through cloning and restriction enzyme analysis of the PCR- amplified fragments showing unique RFLP pattern, 6 novel cry 1-type genes were identified and named cry 1-201, cry 1-202, cry 1-203, cry 1-205, cry 1-206 and cry 1-207, respectively. In the nucleotide sequence analysis of these novel crystal genes, cry 1-201, cry 1-202 and cry 1-203 showed 93.7%, 94.5% and 66.7% of maximum similarities with cry 1 Ac gene, respectively. Also, cry 1-205 and cry 1-207 showed 99.5% and 99.4% maximum similarities with cry 1 Be gene, and cry 1-206 showed 97.5% maximum similarity with cry 1 Ab gene.
