Abstract

Fingerprinting identification and cluster analysis were conducted among 9 introduced sugar beet (B. vulgaris) cultivars from Holland and 4 lines from China using Inter simple sequence repeats (ISSR) markers. Six stable and high polymorphic primes were selected in this study. 48 bands were obtained using ISSR --PCR with the six primers, and polymorphic rate was 87.5% among the thirteen sugar beet cutivars and lines. All test varieties can be distinguished by ISSR fingerprinting with the six primes. Fingerprinting map and digit identification code were constructed by 8 polymorphic bands from primer UBC813 and L1, which can discriminate 13 cultivars (lines) completely. The result indicated that ISSR fingerprinting could identify different sugar beet varieties efficiently. The genetic similarity matrix among all 13 sugar beet varieties was obtained by similarity analysis using Dice algorithm with NTSYS-pc software. Similarity coefficients ranged from 0.46 to 0.89 with an average of 0.67. Cluster tree was formed using unweighted pair-group method with arithmetic means (UPGMA) method. Cluster analysis showed 13 varieties (lines) were divided four groups and two subgroups. UPGMA cluster analysis showed clear genetic relationships among the 13 sugar beet cultivars. The result indicated that the relationship of sugar beet materials was connected with geography factor and character of breeding and ISSR markers can be used to estimate genetic distance of plant.
