Abstract

Summary Yellow fever (YF) virus strain 17D-infected and- uninfected SW13 cells were labelled with 3H-uridine in the presence of actinomycin D. A maximum of viral specific incorporation of label was observed between the 25th and 27th of the viral cycle. Viral RNA molecules were analysed by SDS-sucrose gradient centrifugation and agarose gel electrophoresis. At least three classes of RNA with sedimentation coefficients of 40, 26 and 20 S wee observed in cytoplasmic extracts from infected cells. The only RNA species associated with polysomes was the genome size 40-S RNA molecule; the 20-S RNA species appeared to be a replicative form composed of a positive and a negative 40-S RNA molecule; the 26-S RNA species probably corresponded to a replicative intermediate.
