Abstract

Objective To investigate hypoxia-inducible factor-1α ( HIF-1α), cyclooxygenase-2 (COX-2) and C-reactive protein (CRP) mRNA transcription level of ulcerative colitis pathological tissues and expression levels of HIF-1α, COX-2 and CRP of ulcerative colitis serum. Methods Ninety-eight patients with ulcerative colitis were divided into group A ( mild), group B (moderate) and group C (severe) according to the severity, with 25 patients of volunteers as the control group D. RT-PCR was used to analyze mRNA levels of HIF-1α, COX-2 and C-reactive protein in ulcerative colitis pathological tissues. ELISA was used to detect the HIF-1α, COX-2 and C-reactive protein levels of serum. Results HIF-1α mRNA levels gradually increased with the exacerbations of ulcerative colitis. COX-2 mRNA levels of group A were signifi- cantly higher than those of Group D ( P 〈 0. 05 ), COX-2 mRNA levels of Group B were significantly higher than of group A (P 〈 0. 05 ), and COX-2 mRNA levels of group C have no significant difference compared with group B ( P 〉 0. 05 ). CRP mRNA levels of group A have no significant difference compared with group D ( P 〉 0. 05 ), CRP mRNA levels of group B were significantly higher than those of group A ( P 〈 0. 05 ), and CRP mRNA of group C were significantly higher than group B ( P 〈 0.05 ). The average serum protein levels of HIF-1α, COX-2 and C-reactive protein gradually increased with the exacerbations of ulcerative coli- tis. Conclusion HIF-1α, COX-2 and C-reactive protein mRNA levels of lesion cells and their serum levels were correlated with the progression of ulcerative colitis positively, suggesting that the detection of HIF-1α,COX-2 and C-reactive protein mRNA levels and expression levels can be used to predict the severity of ulcerative colitis.
