Abstract

Objective To investigate the expression of hypoxia-inducible factor-lα(HIF-1α) and its effect on the proliferation bone marrow mesenchymal stem cells (BMSCs) in lung tissue after paraquat poisoning. Methods All 120 SD rats with paraquat poisoning were randomly divided into three groups.Rats were treated with HIF-1α antisense oligonucleotides (HIF-1α asON)or HIF-1α missense oligonucleotides (HIF-1α msON). The control rats were treated with normal saline.On day1, 4, 8 and 12 after oligonucleotide transfection, peripheral blood bone marrow mesenchymal stem cells (BMSCs), serum vascular endothelial growth factor (VEGF) levels, lung tissue HIF-1α and VEGF protein and mRNA were detected by immunohistochemistry, enzyme-linked immuno assay (ELISA) and PCR. Results On day 1 and day 4 after the treatment HIF-1α asON, the lung tissue HIF-1α, VEGF protein and mRNA, peripheral blood BMSCs, and VEGF levels were significantly lower than that of the rats treated with either HIF-1α msON or saline control (P<0.05). Conclusion The expression of HIF-1α in lung tissue after paraquat poisoning was increased, which promotes the proliferation of BMSCs through VEGF.
