Abstract

Hyperpatulones A (1) and B (2), two new polycyclic polyprenylated acylphloroglucinols (PPAPs), were isolated from the leaves of Hypericum patulum. Their unusual 6,7,5-tricyclic system with a peroxide bridge structures were elucidated by comprehensive analysis of their spectroscopic and HRESIMS data. The absolute configuration of hyperpatulone A was determined by comparison between experimental and calculated electron circular dichroism (ECD) spectra. Compounds 1 and 2 exhibited moderate α-glucosidase inhibitory activity with IC50 values of 81.45 ± 1.04 and 12.81 ± 2.13 μM, respectively. Molecular docking studies of 1 and 2 with human maltase-glucoamylase (MGAM) revealing that 2 showed higher shape complementarity with the binding pocket.
