Abstract

1. 1. It has previously been found that during hydroxyapatite thermal chromatography of mouse-liver chromatin and DNA, mouse-satellite DNA eluted at higher temperature from chromatin, than it did from purified DNA. 2. 2. The elution profile of mouse-satellite DNA in chromatin depleted of histone H1 was shifted to lower temperature. After the removal of all lysine-rich histones (HI + H2A + H2B) a further shift occurred and the elution profile coincided with that obtained upon the melting of purified DNA. 3. 3. The reconstitution of chromatin restored the higher temperature elution profile of satellite DNA.
