Abstract

To further explore the mechanism of secretory cell metaplasia in hamsters induced by human neutrophil elastase (HNE) we investigated the effects of HNE on glycoconjugate release in hamster tracheal ring organ cultures. Tracheal ring organ cultures were established from male hamsters 12–15 weeks of age and 115–130 g in weight. We used Minimal Essential Medium with Earl's salts, antibiotics (50μg/ml streptomycin sulfate, 50 U/ml penicillin G), 1 μM retinoic acid and 5% neonatal bovine serum. One day old cultures were pulsed with 10μCi/ml of 3H-glucosamine (sp act 8Ci/ mmol) for 18 hr.
