Abstract

This study aims to determine the relationship between the intensity of estrus and estradiol concentrations during insemination on local cattle. Ten cows were divided into two groups with each group consist of five cows with category: aged 5-8 years, weight 150-250 kg, clinically healthy, and not pregnant. Plasma samples for examination of the hormone estradiol concentrations were collected during insemination time from jugular vein. Prior to the collection of plasma samples, whole cow were estrus synchronized using prostaglandin F2 alpha (PGF2α) and Ovsynch protocol. The PGF2α were injected twice at intervals of 11 days. Examination of estradiol hormone concentration was performed with enzyme-linked immunosorbent assay (ELISA) using estradiol commercial kits. The results of this study indicate that statistically there is no relationship between estrous intensity and estradiol concentration.

MATERI DAN METODE

Dalam penelitian ini digunakan 10 ekor sapi betina lokal, umur 5-8 tahun, mempunyai berat badan 150-250 kg, betina dengan status tidak bunting, mempunyai dua siklus reguler, dan >90 hari pascapartus, dan sehat secara klinis. Sinkronisasi berahi Pada kelompok I, hari ke-0 dilakukan palpasi rektal untuk mengetahui status reproduksi. Sapi-sapi yang mempunyai status reproduksi baik dan tidak bunting pada hari ke-1 dilakukan penyuntikan pertama PGF2α (2 ml Prostavet C, berisi 5 mg Etiproston yang merupakan PGF2α sintetik). Pada kelompok II, hari ke-1 diawali dengan penyuntikan GnRH (1 ml Fertagyl, yang berisi 0,1 mg/ml Gonadorelin yang merupakan GnRH sintetik). Pada hari ke-7 disuntik dengan PGF2α (2 ml Prostavet C, berisi 5 mg Etiproston yang merupakan PGF2α sintetik). Darah kemudian dibawa ke dalam laboratorium untuk diambil serumnya dan didiamkan selama 30 menit sebelum dilakukan sentrifuga. Pengukuran Konsentrasi Estradiol Pada masing-masing plat ELISA dimasukkan 25 μl larutan standar, sampel, dan kontrol, kemudian dicampur masing-masing dengan 200 μl reagen konjugat estradiol pada masing-masing well selanjutnya inkubasi selama 120 menit pada suhu ruangan.
