Abstract

Orostachys japonicus is traditionally used as anti‐inflammatory, anti‐febrile, hemostatic, anti‐dote, and anti‐cancer agent in Korea. The soluble faction of ethyl acetate (EtOAc) showed the highest anti‐cancer activity on AGS human gastric cancer cells. Further factions of EtOAc extracts were performed by HPLC. The aim of the present study was to investigate the apoptotic activity of further fraction of EtOAc fraction of O. japonicus in AGS cells. ASG cells were cultured in several concentration of further fraction of EtOAc fraction. The apoptotic effect of further fractions of EtOAc fraction was measured using MTS assay, annexin V‐FITC, PI‐labeling, and western blotting. In MTS assay, the survival rates were decreased in dose‐dependent manner. O. japonicus‐induced apoptosis was confirmed by assays with annexin V‐FITC, PI‐labeling. The increase in apoptosis level induced was associated with down‐regulation of Bcl‐2 and activation of Bax, caspase ‐3, ‐8 and ‐9.
