Abstract

Abstract A novel method was proposed to screen and analyze the membrane permeable ingredient group by hollow fiber liposome microscreening (HFLMS) coupled with high performance liquid chromatography (HPLC). In the HFLMS coupled with HPLC, the hollow fiber filled with liposome was used as a screening device, and the membrane permeable ingredient group were screened, fished and isolated from traditional Chinese medicine (TCM) extracts in phosphate buffer (pH 7.4). In this work, the surface nature of the hollow fiber with liposome was characterized, the nonspecific binding between activity enter of hollow fiber and the target compound was discussed, and the reproducibility of HFLMS coupled with HPLC was studied. Using this method, the coumarins and lignans compound groups that could permeate through the liposome membrane were screened and fished from TCM extracts, respectively. Some structures of the lignans and coumarins combined with liposomes in TCMs were identified by the comparison to the retention times of the reference substances.
