Bal and colleagues have now uncovered recurrent mutations in noncoding regions in super-enhancers, leading to loss of transcriptional repression of oncogenes, resulting in tumorigenesis. These recurrent mutations share an activation-induced cytidine deaminase signature implicating activation-induced cytidine deaminase as the mutational driver. CRISPR–Cas9-mediated reversion of these mutations led to a significant decrease in oncogene expression and prevented proliferation and survival of DLBCL cell lines. Expert Commentary: This study has uncovered a previously unrecognized role for activation-induced cytidine deaminase-mediated hypermutation of super-enhancers in lymphomagenesis and suggests potential new drug targets in DLBCL.
