Hagiwara and colleagues profiled 2,860 long-term survivors by deep sequencing and detected clonal hematopoiesis (CH) in 15% of survivors, which is significantly higher than in community controls (8.5%). CH in survivors is associated with exposures to alkylating agents, radiation, and bleomycin and is significantly enriched in TP53 and STAT3. Mutations in STAT3, characterized as a CH gene specific to Hodgkin lymphoma survivors, occur predominantly in T cells and are contributed by SBS25, a mutational signature associated with procarbazine exposure. Longitudinal monitoring is recommended to ascertain the long-term effects of CH in pediatric cancer survivors. See article, p. 844.
