Abstract

Objective To investigate the association of Lipopolysaccharides (LPS), Toll like receptor(TLR), TNF-α and ET-1 with the development of HPS. Methods 71 patients were divided into 3 groups: end-stage liver disease with HPS(HPS group, n=31, 26patients were performed liver transplantation), end-stage liver disease without HPS(non-HPS group, n=30) and healthy volunteers(n=10). The written informed consents of all patients were obtained and the ethical committee approval was received. Blood was collected from these 71patients before and 3, 7, 14, 21 and 28days after OLT to detect TLR2mRNA and iNOSmRNA in peripheral blood monocytes and plasma LPS, TNF-α and ET-1 level. Also their changes after OLT were observed. Results TLR2mRNA, iNOSmRNA, LPS, TNF-α and ET-1 before OLT in HPS group were significantly higher than that in control group(P<0.05).They were also higher than those in non-HPS group but without significance(p>0.05). TLR2mRNA, iNOSmRNA, LPS, TLR, TNF-α before OLT was higher in non-HPS group than those in control group without significance(P>0.05 ), except for significantly higher ET-1 in non-HPS group (P=0.001). TLR2mRNA, TNF-αand ET-1 were all decreased in HPS and non-HPS group after OLT compared with those before OLT. Conclusion Higher lipopolysaccharides at the end stage of liver disease with the release of series of inflammatory factors may be associated with the development of HPS.
