Abstract

Cyclic GMP and the dibutyryl derivative of cyclic AMP, but not cyclic AMP itself, were found to increase release of growth hormone from incubated pituitary explants. Cyclic GMP-induced growth hormone release was potentiated by concomitant treatment of the pituitaries with aminophylline. Hypothalamic extracts containing growth hormone releasing activity increased the intracellular concentration of both cyclic GMP and cyclic AMP in incubated pituitaries. These studies indicate that either cyclic GMP can serve individually as an intracellular mediator of growth hormone release or alternatively that both cyclic GMP and cyclic AMP are necessary to promote growth hormone release.
