Abstract
Glycocalicinis the carbohydrate-richexternalportion of plateletglycoproteinGPIb alpha. The Ieucine-rich45Kdaminoterminalpornonof glycocalicinencompasses295 amino acids and it has beenidentifkxiasthe bindingsiteforthrombin and von Willebrandfactor(vWfJ Vicenteet al (1) showed that vWf binds to glycocalicinwithin the sequence S251-Y279 with nstocetin and botrocetin. Katagiriet al (2) identifiedanothersequence,D235-K262that inhibitedvWfbindingto GPIb alphawithristocetin. In our studiesof glycocalicinand its fragmentswe have identifiedand examinedtherole of a glycocalicinderivedpeptide,G190-L214, in vWf bindingto platelets.We haveconstructedshortened,substitutedand deletedpeptides to Iodize a bindingsite(s) for vWf in thepresenceof nstocetin,botrocetinand asialovWf.
