Abstract

While significant progress has been made in understanding the roles of Akt and NF-κB signaling pathways in oncogenesis and cancer progression, little is known regarding proteins that function in both pathways and their effects on development of tumor. Here, we report that the expression level of GGAP2 was in creased and significantly correlated with that of Akt in cancer cell lines and human prostate cancer patients. GGAP2 interacted with Akt and p50 subunit of NF-κB transcription factor and stimulated their activation. Moreover, GGAP2-expressing human prostate cancer cell lines showed higher malignant transformation ability and significantly enhanced tumor growth in vivo. Our results elucidate a novel mechanism by which Akt and NF-κB pathways promote cancer cell growth synergistically through GGAP2.
