Abstract

The application of gene-therapy strategies for the treatment of cancer has finally been realised in the form of numerous phase I clinical trials. Genetic prodrug activation therapy (GPAT) (figure) uses the transcriptional differences between normal and neoplastic cells to drive the selective expression of a metabolic suicide gene (usually derived from bacteria or viruses) able to convert a non-toxic prodrug to its toxic anabolite.1
