Abstract

The taxonomic classification of mudskipper species is controversial due to their similar body. Five samples of mudskipper fish collected from Bogowonto Lagoon were used for genetic identification and sequence comparison of mitochondrial DNA encoded partial cytochrome oxidase subunit I (COI) gene. Sequence analysis of COI gene identified two species of mudskipper, Periophthalmus argentilineatus and Periophthalmus kalolo. For the 522 bp sequence determined in P. argentilineatus, three haplotypes were revealed with six variable sites. Haplotype diversity of P. argentilineatus was 1.0 and nucleotide diversity was 0.0077. In addition, the genetic divergence varied from 0.2 % to 1.2 %. Phylogenetic analysis showed that all of the individuals of P. argentilineatus sampled from Bogowonto Lagoon clustered together with samples from Pandeglang (West Java, Indonesia) but separated and genetically differentiated with P. argentilineatus from Jembrana (Bali, Indonesia) and China which were previously examined. Furthermore, no intraspecific variation of P. kalolo samples from this study. However, COI gene sequences of P. kalolo from Bogowonto Lagoon were different from the COI sequences of P. kalolo from Cilacap (Central Java, Indonesia) and Tulung Agung (East Java, Indonesia) which were previously investigated.
