Abstract
Objective To analyze the genetic characteristics of measles virus strains causing an outbreak in Aba prefecture of Sichuan province. Methods Eight throat swab specimens from a measles outbreak occurred in a township primary school in Hongyuan county, Aba prefecture, Sichuan province from March 16 to April 3, 2017 were collected. After cultured by Vero/SLAM cells, a 634-bp-fragment on C-terminal of the N gene was amplified by reverse transcription-polymerase chain reaction (RT-PCR) and subjected to sequence and similarity analysis. Results Three strains of measles virus were isolated from 8 throat swab specimens and were all identified as H1a subtype. The nucleotides and amino acids similarity between the isolates and the H1a subtype reference strain - Chin9322 were 98.9% and 99.3%, respectively. The nucleotides similarities between the isolates and the measles virus isolated from other cities in Sichuan Province and Aba Prefecture in 2017 were 99.3%-100.0% and 98.9%-100.0%, respectively. The amino acids similarities were 98.6%-100.0% and 98.0%-100.0%, respectively. Conclusions This outbreak of measles was caused by H1a subtype measles virus.
