Abstract

Background: Parkinson’s disease (PD) is a neurodegenerative disorder characterized by α-synuclein aggregation in the brain and loss of dopaminergic neurons in the substantia nigra. In a genetic analysis by Mandel et al, they found an increased expression of EGLN1, a oxygen/iron sensor, in the substantia nigra of parkinsonian brains. The up regulation of EGLN1 promotes hypoxia inducible factor and iron regulatory protein degradation and leads to iron accumulation, which is known to induce oxidative stress and also promotes α-synuclein aggregation.
